چکیده
Type 1 interferons (IFN-1) are major drivers of disease activity in systemic (SLE) and cutaneous lupus erythematosus (CLE). Plasmacytoid dendritic cells (pDCs) the producers IFN-1 during viral infection. pDCs reduced circulation SLE have recently been shown to an exhausted phenotype, suggesting that pathogenic were located end organs. We performed imaging mass cytometry treatment naïve CLE skin (n=48), which revealed only 8.5% (IQR 0.0-22.22) IFNa+. Across all biopsies, second lowest contributor absolute IFNa+ (Median 1; IQR 0-4.5), with B lymphocytes being smallest contributor. Classic (cDCs) macrophages (Mf) largest relative contributors IFNa CLE. For IFNb, 16.7% positive, compared 60.7% CD14+CD16+ Mf. also IFNb+ 0-7), again lowest. then mRNA situ hybridization for CLEC4C (BDCA2) (a, b, k) genes confirm these findings. was detected ITGAX (CD11c) positive cDCs but we did not identify colocalization mRNA. eluted leukocytes from flow cytometry. Flow showed a median 5.2% 3.8-9.6), other myeloid cell lineages such as CD68+ Mf 35.9% (26.4-40.7), M1 23.7% (21.6-31.7), M2 (26.4-40.7). CyTOF on PBMCs 41-marker panel, including pan-IFNa demonstrated minimal staining lineages. Taken together, findings suggest may play central role larger numbers percent. through IFN-1-independent mechanisms.

Plasmacytoid dendritic cells are present in cutaneous dermatomyositis lesions in a pattern distinct from lupus erythematosus.
BACKGROUND Plasmacytoid dendritic cells (PDCs) are CD123-positive dendritic cells (DCs) capable of producing interferon-alpha (IFN). They are thought to play a role in anti-viral immunity and the pathogenesis of lupus erythematosus (LE). Given the histologic similarities between LE and dermatomyositis (DM), we evaluated the presence and distribution of PDCs in lesional skin of both diseases. ...
متن کاملEBI2 Is a Negative Regulator of Type I Interferons in Plasmacytoid and Myeloid Dendritic Cells
Epstein-Barr virus induced receptor 2 (EBI2), a Gαi-coupled G protein-coupled receptor, is a chemotactic receptor for B, T and dendritic cells (DC). Genetic studies have also implicated EBI2 as a regulator of an interferon regulatory factor 7 (IRF7)-driven inflammatory network (IDIN) associated with autoimmune diseases, although the corollary in primary type I IFN-producing cells has not been r...
متن کاملPlasmacytoid dendritic cells in angiolymphoid hyperplasia with eosinophilia
Background: Angiolymphoid hyperplasia with eosinophilia (ALHE) is characterized by irregularly-shaped blood vessels with an inflammatory infiltrate. While absent from normal skin, plasmacytoid dendritic cells (pDCs) infiltrate the skin upon injury and during several infectious, inflammatory, and neoplastic entities. In addition to providing anti-viral resistance, pDCs link the innate and adapti...
متن کاملPlasmacytoid dendritic cells sense skin injury and promote wound healing through type I interferons
Plasmacytoid dendritic cells (pDCs) are specialized type I interferon (IFN-α/β)-producing cells that express intracellular toll-like receptor (TLR) 7 and TLR9 and recognize viral nucleic acids in the context of infections. We show that pDCs also have the ability to sense host-derived nucleic acids released in common skin wounds. pDCs were found to rapidly infiltrate both murine and human skin w...
متن کاملProduction of interferons by dendritic cells, plasmacytoid cells, natural killer cells, and interferon-producing killer dendritic cells.
The capacity of mouse spleen conventional dendritic cells (cDCs) and plasmacytoid dendritic cells (pDCs) to produce interferon-gamma (IFN-gamma) or IFN-alpha was assessed, and compared with that of natural killer (NK) cells and the recently identified interferon-producing killer dendritic cells (IKDCs), both of which are frequent contaminants in DC preparations. Fully developed cDCs or pDCs, if...
